DnrNはresponse regulatorに配列は似ているが、putative phosphorylation siteの置換(D55E, D55N)をしても活性が完全にはなくならず、近くにHKが見当たらないことからpseudo-response regulatorとも呼ばれている。

dnrN::aphII mutationはdnrIで相補可能。
dnrI::aphII mutantはdnrNで相補できない。
よってdrnNがdnrIの転写を活性化するというregulatory cascadeの存在が示唆される。

DnrNをE.coliで過剰発現し、抽出液でgel mobility shift analysis.
DnrNは高い親和性で、dnrI promoter領域に特異的に結合する。
wild-type DnrNとputative phosphorylation site置換(D55N)mutantで結合能力に違いがなく、DnrNのリン酸化はDNA bindingに必要ない。
dnrN発現がDNR産生と同時に起こり、DnrNのレベルはDNRを産生しないmutant株で低いので、dnrNとdnrIの発現はDNRによってfeedback調節されると推測される。

dnrO::aphII mutant strainはdaunorubicin (DNR)非産生。
このmutant strainにdnrN or dnrI を導入すると生合成可能。
dnrI 発現はDnrNが必要なことがわかっているので、dnrN発現にDnrOが必要ということが示される。

DnrO → dnrN = DnrN → dnrI = DnrI →DNR生合成genes

DnrOをE.coliで過剰発現し、抽出液でmobility shift DNA-binding assays。
DnrOが特異的にoverlapping dnrN/dnrO(p1) promoter regionへ結合するので、DnrOはdnrNとdnrOの両方の発現を調節するかもしれない。
