Composition
Tripticase peptone 5.00 g Peptone 5.00 g Bacto Yeast Extract (Difco) 10.00 g Beef extract 5.00 g Glucose 5.00 g Tween 80 1.00 ml K2HPO4 2.00 g Sal Solution* 40.00 ml Vitamin K1 solution** 0.20 ml Hemin solution*** 10.00 ml Resazurin 1 mg Cysteine-HCl 0.5 g Distilled water 950 ml Mix ingredients except for Vitamin K1 solution. Dispense the medium into suitable culture vessels and autoclave under a N2 atmosphere. Sterilize vitamin solution by filatration and add to the medium after autoclave. *Salt Solution MgSO4·7H2O 0.25 g CaCl2·2H2O 0.50 g K2HPO4 1.00 g KH2PO4 1.00 g NaCl 2.00 g NaHCO3 10.00 g Distilled water 1 L **Vitamin K1 solution vitamin K1 0.10 ml 95% Ethanol 20.00 ml Mix ingredients and filter sterilize. Refrigerate the solution in the dark place.   ***Hemin solution Hemin 50 mg 1 N NaOH 1 ml Distilled water 100 ml Dissolve hemin in 1 ml of 1 N NaOH. Adjust volume to 100 ml with distilled water.
