Prepare filter sterilized 0.5 M CaCl2 and 0.6 M MgCl2 solution first.
Fill up 5ml of 0.5 M HEPES to 100 ml with UPW and adjust pH to 7.4, then sterilized by autoclaving. After autoclaving, add 600 μl of 0.5 M CaCl2 and 333 µl of 0.6 M MgCl2.
