CrpG: amino acid decarboxylase

ラベリング実験などでmethyl beta-alanineがmethyl-aspartateから形成されるかどうかを検討。
crptophycin1のunitCであるmethyl-beta-alanineの2R-stereoisomerを産出するため、CrpGはR-methyl aspartateに対して高い選択性を示す。

CrpGをクローニング、過剰発現、精製、活性確認。
(2S,3R)-3-methylaspartic acidをdecarboxylateして、3-amino-2(R)-methylpropionic acidを形成するbeta-methylaspartate-alpha-decarboxylaseである。
3-amino-2(R)-methylpropionic acidはcryptophycinsのUnit Cに取り込まれる。
