MxaF: PKS domains (ACP-L, KS, AT-L, AT-1, DH, KR, ACP2)

mxaFはloading + module 1をコード。
mxaFのinactivationでmyxalamidsは産生されなくなる。

module 1 AT domainは2つある。
1つめのAT domain(AT-L)はmyxalamid A-Dの生合成のために異なるstarter unitを認識する。

myxalamid A: 2-methylbutyrate
myxalamid B: isobutyrate
myxalamid C: propionate
myxalamid D: acetate

これらを結合するのはloading moduleのACP-L.

Myxococcus xanthus由来のmyxalamid生合成gene clusterの同定論文。

M. xanthus and Stigmatella aurantiacaのbkd mutantsからnew myxalamidsとmyxalamid Bを検出。
bkdは、isoleucine and valineから、myxalamid A and Bのstarter unitである2-methyl-butyryl-CoA and isobutyryl-CoAへの触媒をするbranched-chain ketoacid dehydrogenaseをコードする。

この結果からbkd mutantでのisobutyryl-CoAの起源を調査。
M. xanthus bkd mutantではvalineから生じるが、S. aurantiacaではiso-odd fatty acidsのalpha-oxidationに由来する。
