cytochrome P450候補ORF4つ(ORF4, 5, 13, 16)のinactivation mutantを作成し、表現型を解析。

orf5 mutantはrifamycin B産生が抑制され、中間体であるrifamycin Wが蓄積した。また、cytochrome P450 monooxygenases inhibitorであるancymidolで処理したwild株の二次代謝産物をTLC, HPLC and ESI-MS analysesで解析し、やはりrifamycin Wを蓄積していることを確認。
よってRif-Orf5はrifamycin Wからの変換を担うcytochrome P450である。

この結果をふまえ、Orf5はrifamycin WのC12-C29二重結合を酸化的切断し、ketal構造を形成するための再構築反応を導くと提唱。
