rubK, rubL, rubM, and rubNはsingle operon。

rubN以外がAHBA合成rif genesに高い相同性があると言われているが、RubN vs. RifNも他のものに比べれば劣るがかなり有意に相同性がある(id49%)。

rifamycinと関連ansamycinsのPKS starter 3-amino-5-hydroxybenzoic acid (AHBA)の生合成経路について調査。関連遺伝子の不活化、異種性発現、中間体を基質とした反応確認など。

どちらもRifN: kinase

rifNは、PKSのstarter unitであるAHBAの合成に必要十分な7genes(rifG, H, J, K, L, M, N)のひとつだが、rifH, G, Jと異なりshikimate pathway関連遺伝子には似ていない。

rifN mutantの実験から、AHBA生合成の早期段階で作用し、aminoDAHP形成か、それより前で関与することが示唆されている。

rifNについての実験論文。

E.coliでHis6 fusion proteinとして過剰発現、精製したRifNの基質特異性を様々な糖で確認。kanosamineのみが反応し、この反応生成物をNMRなどで解析したところ、kanosamine 6-phosphateと同定された。

よって、RifNはkanosamine kinase
