rubransarolのmorpholine環形成のための様式は不明なままだが、rubF4 to rubF7 and rubP3 genesがこの過程で役割を果たすかもしれない。

aminocoumarin生合成、結合に関連するputative acyladenylate-forming enzymesの候補、Orf4, RubF6, RubC1を発現、精製して調査している。
RubF6 (56 kDa)は不活性だが、site-directed mutagenesisにより活性型酵素へ変換されうる。

RubF6はOrf4に高い相同性があることから、これらは遺伝子重複に由来し、その後一方が不活性型へ変異された可能性がある。

3-dimethylallyl-4-hydroxybenzoic acid (ring A of novobiocin) と
3-amino-4,7-dihydroxy-8-methyl coumarin (ring B of novobiocin)
の間のアミド結合の形成をATP-dependent reactionで触媒した。
また、ring A and ATPからのacyl adenylateの形成がATP-PP(i) exchange assayによって実証された。
すなわちNovLは、acyl adenylateのようなring Aの活性化と、引き続くacyl基のring Bのamino基への移動の両方を触媒した。
