VinK (327aa) : acyl CoA-ACP transacylase

配列解析からの機能推定。
type II fatty acid synthasesのmalonyl transferaseに相同性あり。
よってstarter acyl unitをACPに移すことを担うと思われる。

VinJ, K, L, M, N, Oの酵素的解析を行っている論文。

VinP1のACP domainのみを発現し、Sfpでholo型に変換。
alanine-3-amino-2-methylpropionyl-VinL と VinP1-ACPをVinKで反応させ、産物をLC-ESI-MS解析。効率的なdipeptide transferが起こり、alanine-3-amino-2-methylpropionyl-VinP1-ACPが形成された。

また、VinKの基質特異性を確認するため、3つのaminoacyl-VinLs
・3-methylaspartate-VinL
・3-amino-2-methylpropionyl-VinL
・alanine-3-amino-2-methylpropionyl-VinL
を基質として検討。

alanine-3-amino-2-methylpropionyl-VinLのみが選択的にVinP1-ACPへ移動された。
