AviSは、さまざまなNDP-hexose 2,3-dehydratasesと相同性を示す。
AviS,AviT,AviZ3はNDP-4-keto-6-deoxy-D-glucose→NDP-D-olivoseの反応に関与すると推測している。

また、この反応の中間体NDP-4-keto-2,6-dideoxy-D-glucoseがD-olivoseのC3位がメチル化されている2-Deoxy-D-evalose生合成経路への分岐点であると予測している。

明らかに酵素反応は不安定な化合物を産生しており、たぶんdTDP-3,4-diketo-2,6-dideoxyglucoseまたはその2,3-enolで、dTDPの容易な除去を起こす。

Gra-orf26 + dTDP-4-keto-6-deoxyglucose + NAD(P)H → 反応なし
Gra-orf26 + dTDP-4-keto-6-deoxyglucose + NAD(P)H + Gra-orf27 → dTDP-4-keto-2,6-dideoxyglucose

Gra-orf27をTu99-orf10で、Gra-orf26をTu99-orf11で置き換えても同じ産物を得た。(Tu99 genesはoleandomycin生合成genes)
NADPHがbetterな還元剤だが、NADHでも代わりできる。

Gra-orf27 または Tu99-orf10は、dTDP-4-keto-6-deoxyglucose 2,3-dehydratasesとして働き、
dTDP-4-keto-6-deoxyglucose → dTDP-3,4-diketo-2,6-dideoxyglucoseへ変換する。
dTDP-3,4-diketo-2,6-dideoxyglucoseは3-ketoreductasesであるGra-orf26 または Tu99-orf11 によって還元的に捕獲され、dTDP-4-keto-2,6-dideoxyglucoseを与える。

SpnOはforosamine合成の初期段階でNDP-4-keto-6-deoxy-glucoseの2,3位の脱水反応を行う。

dTDP-4-keto-6-deoxy-D-glucose 2,3-dehydratase_SpnO

dTDP-D-glucose, RfbB(dTDP-D-glucose→dTDP-4-keto-6-deoxy-D-glucose), and either NADH or NADPHと一緒にSpnO and SpnNをインキュベートし、NAD(P)Hの消費量をフォローしてtandem reactionを測定。SpnO or SpnNのどちらかを欠いた場合、消費が見られなかった。
dTDP-3,4-diketo-2,6-dideoxy-D-glucoseは不安定で急速にdTDP and maltol に分解してしまうため、SpnOのみの触媒効率は定量できていない。
