branched-chain alpha-keto acid dehydrogenase (BCDH) complex bkdABの機能解析報告。

アミノ酸配列の相同性と酵素活性により、BkdAはE1 alpha,beta subunitのfusionタイプであることが示された。

bkdA欠損株を作製。
virginiamycin Mの生産能を欠いていたが、virginiamycin Sの生産には影響がなかった。
相補実験により、virginiamycin Mの生産能を回復したことを確認済み。
bkd geneは細胞の生育や形態分化には関与していなかった。
宿主大腸菌でのbkdA過剰発現株作製し触媒活性を測定、bkdAがalpha beta subunitの融合形成をコードする事を確認。
以上より、bkdAB clusterは、virginiamycin Mの生産に必須であることを示した。

Virginiamycin M産生におけるスターターユニットisobutyryl-CoAの提供をプライマーユニットとして経由しているのではないかと予測している。
