Azinomycin B生合成遺伝子クラスターの報告。

aziB2: O-methyltransferase

配列解析に基づき、azinomycin B骨格組み立ての最初のbuilding blockである3-methoxy-5-methyl-naphthoic acid(NPA)は、5-methyl-NPAがAziB1(putative P450 hydroxylase)によりC3でOH基を導入し、続いてAziB2(putative O-methyltransferase)によりメチル化されてできると考えられた。

S.albusでaziBのみを異種発現させると5-methyl-NPAが、aziB, B1, B2を発現させると3-methoxy-5-methyl-NPAが産生された。

この結果からaziB1, aziB2の機能が確認され、azinomycin B骨格を組み立てるNRPS AziA1の基質として3-methoxy-5-methyl-NPAが強く支持された。

AziB2がSAMを利用して3-hydroxyl-5-methyl-NPAから3-methoxy-5-methyl-NPAを形成することを複数の実験で確認している。
