シークエンスされた領域で、MonACPX(polyketide鎖放出に関連する)に相当する分離したACPの候補はorf18のみ。

orf18不活化mutantはwild-typeと比べてsalinomycin産生が50-60%低下した。
よって、Orf18はsalinomycin生合成に関連しているかもしれないが、polyketide鎖放出に必須ではない。

glyceryl-S-ACP形成ではなく、palmitoyl-S-ACP形成に関与するACPであることが確認されている。

palmitic acid, CoA, ATP, MgCl2, RkA(palmitoyl-CoA synthetase)を含む混合液にholo-RkBを加えると、palmitic acid → palmitoyl-S-RkBへの実質的変換(約80%)が起こった。

精製されたpalmitoyl-S-RkBを用いて、PKS RkCとの反応も見ている。

daptomycin生合成において脂質化に関与すると考えられているdptEとdptFのクローニング、異種性発現、精製したタンパクの生化学的特徴づけ。

DptEはATPを消費して中〜長鎖分枝脂肪酸を優先的に活性化してacyl-AMPとし、これをCoAではなく、他のACPより特異的にDptF(同族のACP)へ移すことが確認されている。
