17kb fragment(pieA4-B2)を置換したmutantは、Piericidin A1の産生を完全に廃止した。

mutant module 8-TE0(S148A)ではshunt産物としてalpha-pyrone産物が形成されるが、wild-type TE domainを持つmodule 8ではほとんどない。したがって、TE domainはPieA由来beta, delta-diketo thioesterを効率的に加水分解することにより、shunt産物のようなpyronesの形成を基本的に妨げる。

pikromycin TEでmacrolactone形成を方向付ける親水性バリア領域での側鎖Gln183は、Pie TEでは疎水性のLeu107によって置換されているので、その機能性が減っているかもしれない。

macrolactone形成TEと加水分解性TEの機能的な違いは、相対的に限定された基質容器のせいであると提唱されており、Pie TEでも同ケースかもしれない。
